Quantitation comparison of total RNA using the Agilent 2100 bioanalyzer, ribo- green analysis and UV spectrometry Application

Advances in microarray technology have led to the increased demand for techniques appropriate for the analysis of RNA. The two main parameters to be assessed are RNA quality and the quantitation of RNA. Absorbance and fluorescence based measurements are established techniques for RNA quantitation. A new technique for the measurement of RNA is the Agilent 2100 bioanalyzer in combination with the RNA 6000 Nano LabChip® kit. This method offers the advantage of assessing RNA integrity, in addition to a quantitive measurement. This Application Note investigates the various techniques for RNA quantitation. The Agilent 2100 bioanalyzer is compared with UV spectrometry, and the Ribogreen® quantitation system accessing the linear dynamic range, reproducibility, and the effects of contaminants for each quantitation method.
